Effect of Adrenochrome on Calcium Transport and Adenosine Triphosphatase Activities of the Rabbit Sarcoplasmic Reticulum Vesicles

The effect of adrenochrome(5 X 10-6M5 X 10-¢¥M) on Cat+-stimulated Mgt+-dependent adenosine triphosphatase(Caz+-ATPase) and Ca uptake was studied in the sarcoplasmic reticulum(SR) vesicles obtained from rabbit ventricle. Adrenochrome decreased both Caz+-ATPase and Cat+ uptake, although it had the stronger effect on Caz+ uptake than Cat+ -ATPase showing I50 values of 3.16X 10-SM and 10-¢¥M, respectively. The inhibitory effect of adrenochrome on Ca- uptake in SR, vesicles was independent of incubation time. Kinetic study of Caz+ uptake activity at different concentrations of ATP showed that the inhibition was a mixed type. Adrenochrome decreased the activation energy on Caz+-ATPase activity at temperature above 2510. And adrenochrome increased lipid peroxidation of SR vesicles in a dose dependent manner. These results suggest that cardiodepressant effect of adrenochrome may partly be explained on the basis of its inhibitory effect on the calcium trans-porting ability of the SR by the alteration of membrane phospholipid as well as the uncoupling of Caz+ pump mechanism.
